Traditionally, SLAS Discovery has focused on publishing original research papers and reviews. With the introduction of this latest special issue, the journal is expanding its scope to include protocol articles. These articles are designed to provide a detailed, step-by-step guide to a particular experimental technique or methodology, enabling other researchers to replicate the work and build upon it. In this episode, we spoke with one of the special issue Guest Editors, Adam Zweifach, Ph.D. (University of Connecticut), to explore the benefits of protocol articles, how they differ from traditional research papers, and the impact they could have on the field of life drug discovery and greater SLAS scientific community. SLAS Discovery Editor-in-Chief Bob Campbell (Twentyeight-seven Therapeutics) welcomes Sarine Markossian, Ph.D. (National Center for Advancing Translational Sciences (NCATS)) and G. Sitta Sittampalam, Ph.D. (National Center for Advancing Translational Sciences (NCATS)) to discuss the research featured in the latest Special Issue of SLAS Discovery. Titled “Assay Guidance Manual for Drug Discovery: Robust or Go Bust”, this issue showcases the incorporation of best practices in early research stages to improve the efficiency and reliability of the drug discovery process. Much of the research featured in this issue is authored by instructors of the AGM workshops: Assay Guidance Workshops for High-throughput Screening and Lead Discovery. In this Special Issue of SLAS Discovery, the authors explore multiple aspects of cellular target engagement; from direct measurement of compound-target interaction using BRET technologies, through measurement of compound induced changes in protein stability and localisation, to in vivo drug distribution and target engagement studies. They also highlight technologies that enable a more holistic view of target engagement by mapping multiple compound-protein interactions in a single study. When applied in combination, these methods can build a strong platform of evidence linking compound to target to biological response, thereby impacting project attrition and medicines discovery success.
